Novel Hyperactive Transposons for Genetic Modification of Induced Pluripotent and Adult Stem Cells: A Nonviral Paradigm for Coaxed Differentiation 
Researchers have previously generated hyperactive transposases derived from Sleeping Beauty, using an in vitro molecular evolution and selection paradigm. They now demonstrate that these hyperactive transposases resulted in superior gene transfer efficiencies and expression in mesenchymal and muscle stem/progenitor cells, consistent with higher expression levels of therapeutically relevant proteins including coagulation factor IX. Human Gingiva-Derived Mesenchymal Stem Cells Elicit Polarization of M2 Macrophages and Enhance Cutaneous Wound Healing
The findings provide first evidence that mesenchymal stem cells from human gingiva are capable to elicit M2 polarization of macrophages, which might contribute to a marked acceleration of wound healing. [Stem Cells]

Antibacterial Effect of Human Mesenchymal Stem Cells is Mediated in Part from Secretion of the Antimicrobial Peptide LL-37
Researchers studied the effect of human mesenchymal stem cells (MSC) derived from bone marrow on the bacterial growth of gram-negative (E. coli and P. aeruginosa) and gram-positive (S. aureus ) bacteria. MSC as well as their conditioned medium demonstrated marked inhibition of bacterial growth in comparison to control medium or normal human lung fibroblasts. [Stem Cells]

Gold Nanoparticles Promote Osteogenic Differentiation of Mesenchymal Stem Cells Through p38 MAPK Pathway
The results showed that gold nanoparticles promoted the differentiation of mesenchymal stem cells toward osteoblast cells over adipocyte cells by inducing an enhanced osteogenic transcriptional profile and an attenuated adipogenic transcriptional profile. [ACS Nano]

Role of Canonical Wnt Signaling/Beta-Catenin via Dermo1 in Cranial Dermal Cell Development
Cranial dermis develops from cephalic mesoderm and neural crest cells, but what signal(s) specifies the dermal lineage is unclear. Using genetic tools to fate map and manipulate a cranial mesenchymal progenitor population in the supraorbital region, researchers show that the dermal progenitor cells beneath the surface ectoderm process canonical Wnt signaling at the time of specification. Umbilical Cord-Derived Mesenchymal Stromal Cells Modulate Monocyte Function to Suppress T Cell Proliferation
In this paper, researchers report that suppression of mitogen-induced T cell proliferation by human umbilical cord-, bone marrow-, and cord blood-mesenchymal stromal cells required monocytes. [J Immunol]

ARTEMIS Stabilizes the Genome and Modulates Proliferative Responses in Multipotent Mesenchymal Cells
Here, researchers investigate roles for the nonhomologous end joining factor ARTEMIS in multipotent mesenchymal stem/progenitor cells, as putative sarcomagenic cells of origin. [BMC Biol]

Transient Expression of OCT4 is Sufficient to Allow Human Keratinocytes to Change Their Differentiation Pathway
Researchers transiently transfected freshly isolated human skin keratinocytes with the single transcription factor OCT4. Within two days these cells displayed expression of endogenous embryonic genes and showed reduced genomic methylation. More importantly, these cells could be specifically converted into neuronal and contractile mesenchymal cell types. [Gene Ther]

Mesenchymal Stem Cell Mechanics from the Attached to the Suspended State
Human mesenchymal stem cells (hMSCs) are therapeutically useful cells that are typically expanded in vitro on stiff substrata before reimplantation. Here, researchers explore MSC mechanical and structural changes via atomic force microscopy and optical stretching during extended passaging, and demonstrate that cytoskeletal organization and mechanical stiffness of attached MSC populations are strongly modulated over >15 population doublings in vitro. Amniotic Mesenchymal Stem Cells Enhance Normal Fetal Wound Healing
Researchers conclude that fetal wound healing encompasses an autologous yet exogenous cellular component in naturally occurring amniotic mesenchymal stem cells. While seemingly not absolutely essential to the healing process, amniotic cells expedite wound closure and enhance its extracellular matrix profile. [Stem Cells Dev]

Analysis of the Chondrogenic Potential and Secretome of Mesenchymal Stem Cells Derived from Human Umbilical Cord Stroma
Mesenchymal stem cells (MSCs) from umbilical cord stroma were isolated by plastic adherence and characterized by flow cytometry, looking for cells positive for STRO1, OCT3/4 and SSEA-4 as well as the classic MSC-markers CD44, CD73, CD90, Ki67, CD105 and CD106 and negative for CD34 and CD45.
