TOP STORY
Mechanical Memory and Dosing Influence Stem Cell Fate
Researchers found that the activation of the Yes-associated protein and transcriptional coactivator with PDZ-binding domain as well as the pre-osteogenic transcription factor RUNX2 in human mesenchymal stem cells cultured on soft poly(ethylene glycol) hydrogels depended on previous culture time on stiff tissue culture polystyrene. Comparative Analysis of Mouse Induced Pluripotent Stem Cells and Mesenchymal Stem Cells during Osteogenic Differentiation In Vitro
Scientists provide a direct in vitro comparison of the osteogenic differentiation process in mesenchymal stem cells and induced pluripotent stem cells from adult C57BL/6J mice. [Stem Cells Dev] Abstract

Functional Comparison of Human Induced Pluripotent Stem Cell-Derived Mesenchymal Cells and Bone Marrow-Derived Mesenchymal Stromal Cells from the Same Donor
Scientists have derived mesenchymal stem cell-like cells (iMPCs) using four different methodologies from a newly established induced pluripotent stem cell line reprogrammed from human bone marrow stromal cells (BMSCs), and compared the iMPCs directly to the originating parental BMSCs. [Stem Cells Dev] Abstract

Development of a Dynamic Stem Cell Culture Platform for Mesenchymal Stem Cell Adhesion and Evaluation
The authors have developed a device, the dynamic stem cell culture platform, consisting of a biomimetic scaffold cultured within the dynamic environment of a perfusion bioreactor. By varying scaffold parameters including stiffness and protein inclusion at the material surface, they found that mesenchymal stem cells were able to adhere to modified substrates, while still maintaining multipotency. [Mol Pharm] Abstract

Growth Differentiation Factor 6 and Transforming Growth Factor-Beta Differentially Mediate Mesenchymal Stem Cell Differentiation, Composition and Micromechanical Properties of Nucleus Pulposus Constructs
Researchers investigated the effect of growth differentiation factor-6, compared to other transforming growth factor superfamily members, on discogenic differentiation of mesenchymal stem cells, the matrix composition and micromechanics of engineered nucleus pulposus tissue constructs. [Arthritis Res Ther]

Nanotopography Directs Mesenchymal Stem Cells to Osteoblast Lineage through Regulation of MicroRNA-SMAD-BMP-2 Circuit
Researchers investigated if chemically produced nanotopography on titanium surface induces osteoblast differentiation of cultured human bone marrow mesenchymal stem cells by regulating the expression of microRNAs. [J Cell Physiol] Abstract

Mesenchymal Stem Cell or Chondrocyte Seeded Microcarriers as Building Blocks for Cartilage Tissue Engineering
Researchers tested the use of mesenchymal stromal/stem cell- or chondrocyte laden microcarriers as building blocks for engineered cartilage tissue. [Tissue Eng Part A] Abstract

Biochemical and Structural Characterization of Neocartilage Formed by Mesenchymal Stem Cells in Alginate Hydrogels
Investigators focused on mesenchymal stem cells immobilized in alginate hydrogels, and used immunohistochemistry and gene expression analysis combined with advanced microscopy techniques to describe properties of cells and distribution and organization of the forming extracellular matrix. [PLoS One]

Cell-Autonomous Heparanase Modulates Self-Renewal and Migration in Bone Marrow-Derived Mesenchymal Stem Cells
Scientists inhibited the enzymatic activity of cell-autonomous heparanase (HPSE1) in bone marrow-derived mesenchymal stem cells to clarify the physiological role of HPSE1 in bone marrow-derived mesenchymal stem cells. [J Biomed Sci] Abstract

IKVAV Regulates ERK1/2 and Akt Signaling Pathways in BMMSC Population Growth and Proliferation
The molecular mechanism of bone marrow mesenchymal stem cell (BMMSC) population growth and proliferation, induced by Isoleucyl-lysyl-valyl-alanyl-valine (IKVAV), was explored in this study. [Cell Prolif]

REVIEWS
Mesenchymal Stem Cells, Cancer Challenges and New Directions
The authors focus on the therapeutic potential of the human mesenchymal stem cells (hMSC) with special focus on their use in cancer therapy. They consider different mechanisms, by which stem cells can maintain telomeres and thereby the cell’s ability to be expanded in vitro, and also focus on a new therapeutic venue that utilizes hMSCs as delivery vehicles in innovative new cancer treatments. [Eur J Cancer] Abstract

Significance of Epigenetic Landscape in Cartilage Regeneration from the Cartilage Development and Pathology Perspective
The authors compiled and reorganized the existing literature pertaining to epigenetic remodeling events placing emphasis on perceiving the landscape of epigenetic mechanisms during cartilage regeneration with the new operational definition, especially from the environmental factors’ point of view. [Stem Cells Dev] Abstract

Exosomes Derived from Mesenchymal Stem Cells
Compared with cells, exosomes are more stable and reservable, have no risk of aneuploidy, a lower possibility of immune rejection following in vivo allogeneic administration, and may provide an alternative therapy for various diseases. In this review, the authors summarize the properties and biological functions of mesenchymal stem cell-derived exosomes and discuss the related mechanisms.
