In Vitro-Conditioned Bone Marrow-Derived Mesenchymal Stem Cells (BMSC) Promote De Novo Functional Enteric Nerve Regeneration, but Not through Direct-Transdifferentiation
Scientists, using the BAC-induced rat gastric denervation model, aimed to determine whether transplantation of BMSC could promote enteric nervous system neuron regeneration and if so, to elucidate the mechanism. [Stem Cells] Abstract

Perivascular Mesenchymal Stem Cells from the Adult Human Brain Harbor No Instrinsic Neuroectodermal but High Mesodermal Differentiation Potential
Investigators systematically characterized adult human brain-derived pericytes during in vitro expansion and differentiation and compared these cells with fetal and adult human brain-derived neural stem cells and adult human bone marrow-derived mesenchymal stem cells. [Stem Cells Transl Med] Abstract

Health Span-Extending Activity of Human Amniotic Membrane- and Adipose Tissue-Derived Stem Cells in F344 Rats
Human amniotic membrane-derived mesenchymal stem cells (AMMSCs) or adipose tissue-derived mesenchymal stem cells (ADMSCs) were intravenously transplanted to 10-month-old male F344 rats once a month throughout their lives. Transplantation of AMMSCs and ADMSCs improved cognitive and physical functions of naturally aging rats, extending life span by 23.4% and 31.3%, respectively. [Stem Cells Transl Med] Abstract | Press Release

Bile Acids Induce Hepatic Differentiation of Mesenchymal Stem Cells
Since bile acids can promote liver regeneration, their influence on liver-resident and bone marrow-derived mesenchymal stem cells (MSC) was investigated. Physiological concentrations of bile acids such as tauroursodeoxycholic acid were able to initiate hepatic differentiation of MSC via the farnesoid X receptor and transmembrane G-protein-coupled bile acid receptor 5 as investigated with knockout mice. Interleukin-25 Mediates Transcriptional Control of PD-L1 via STAT3 in Multipotent Human Mesenchymal Stromal Cells (hMSCs) to Suppress Th17 Responses
Scientists found that, along with Th1 responses, hMSCs strongly suppressed interleukin-17A (IL-17A)-secreting T (Th17) responses and this required both IL-25—also known as IL-17E—as well as programmed death ligand-1 (PD-L1), a potent cell surface ligand for tolerance induction. [Stem Cell Rep] Full Article | Graphical Abstract

Isolation, Selection and Culture Methods to Enhance Clonogenicity of Mouse Bone Marrow Derived Mesenchymal Stromal Cell Precursors
The authors aimed to optimize the isolation, culture conditions and selection of mouse bone marrow mesenchymal stromal cells. [Stem Cell Res Ther] Full Article

Human Amniotic Mesenchymal Stromal Cells (hAMSCs) as Potential Vehicles for Drug Delivery in Cancer Therapy: An In Vitro Study
The authors investigated mesenchymal stromal cells isolated from the amniotic membrane of human term placenta as candidates for drug delivery in vitro. [Stem Cell Res Ther] Full Article

Human Tubal-Derived Mesenchymal Stromal Cells Associated with Low Level Laser Therapy Significantly Reduces Cigarette Smoke–Induced COPD in C57BL/6 Mice
Scientists studied the effectiveness of human tube derived mesenchymal stromal cells cell therapy associated with low level laser irradiation in experimental cigarette smoke-induced chronic obstructive pulmonary disease. [PLoS One] Full Article

Human Schwann-Like Cells from Adipose-Derived Mesenchymal Stem Cells Rapidly De-Differentiate in the Absence of Stimulating Media
Researchers examined the stability of differentiated adipose-derived stem cells (dASC) by withdrawing differentiation media for 72 hours after the full 18-day differentiation protocol, and measuring changes in morphology, gene expression and protein levels. Withdrawal of differentiation media from dASC resulted in a rapid reversion to stem cell-like characteristics. [Eur J Neurosci] Abstract

Role of Suppressor of Cytokine Signaling 3 in the Immune Modulation of Mesenchymal Stromal Cells
The authors showed that the suppressor of cytokine signaling 3 is an important immune modulator expressed by mesenchymal stromal cells, which is significantly increased by interferon-γ. [Inflammation] Abstract

Type I and II Diabetic Adipose-Derived Stem Cells Respond In Vitro to Dehydrated Human Amnion/Chorion Membrane Allograft Treatment by Increasing Proliferation, Migration, and Altering Cytokine Secretion
Dehydrated human amnion/chorion membrane allografts were evaluated for their ability to regulate diabetic stem cells in vitro. [Adv Wound Care] Abstract | Press Release

Process Development Considerations for Cell Therapy
The authors present an introduction to the process development challenges of cell therapies and describe some of the tools available to address production issues. They provide a summary of what should be considered to efficiently advance a cellular therapy from the research stage through clinical trials and finally toward commercialization. [Stem Cells Transl Med] Abstract
