Pkcδ Is Required for Jagged-1 Induction of hMSC Osteogenic Differentiation
Overexpression of the notch-2 intracellular domain is sufficient to mimic the effect of Jagged-1 ligand (Jag1) on human mesenchymal stem cells (hMSC) osteoblastogenesis, while blocking Notch signaling with a gamma-secretase inhibitor or with dominant negative mastermind inhibits Jag1 induced hMSC osteoblastogenesis. In pursuit of interacting signaling pathways, scientists discovered that treatment with a PKCδ inhibitor abrogates Jag1 induced hMSC osteoblastogenesis. [Stem Cells] Abstract | Press Release

Shockwaves Induce Osteogenic Differentiation of Human Mesenchymal Stem Cells through ATP Release and Activation of P2X7 Receptors
Shockwave treatment promotes bone healing of non-union fracture. Researchers investigated whether this effect could be due to ATP release-induced differentiation of human mesenchymal stem cells (hMSCs) into osteoprogenitor cells. Platelet Lysate Coating on Scaffolds Enhances Directly and Indirectly Cell Migration Improving Bone and Blood Vessel Formation
Investigators aimed to determine if the delivery of human mesenchymal stromal cells (hMSC) seeded on human platelet lysate-coated HA/βTCP scaffolds could enhance vascularization and bone formation, as well as to investigate the mechanisms by which hMSC participate to tissue regeneration. Prospective Isolation of Mesenchymal Stem Cells from Human Bone Marrow Using Novel Antibodies Directed against Sushi Domain Containing 2 (SUSD2)
Recently, the authors described the monoclonal antibodies W3D5 and W5C5, which selectively react with Bone Marrow derived Mesenchymal Stem/Stromal Cells (BM-MSCs) but not with hematopoietic cells. Both antibodies showed an identical reactivity pattern, indicating that they may recognize the same molecule. To identify the cognate antigen, cultured MSCs were sorted for cells expressing either very high levels of W5C5/W3D5 antigen or for cells which were negative for this antigen. Researchers investigated the status of Lectin-like oxidized low density lipoprotein (LDL) receptor-1 expression in bone marrow-derived mesenchymal stem cells (bmMSCs) and its function on bmMSC proliferation. [Exp Cell Res] Abstract

Effects of Salinomycin on Human Bone Marrow-Derived Mesenchymal Stem Cells In Vitro
Various hypotheses on the origin of cancer stem cells (CSCs) exist, including that CSCs develop from transformed human bone marrow mesenchymal stem cells (hBMSC). Since the polyether antibiotic salinomycin selectively kills CSCs, the authors aimed to elucidate the effects of salinomycin on normal hBMSC. [Toxicol Lett] Abstract

Human Adipose Tissue-Derived Mesenchymal Stem Cells Improve Cognitive Function and Physical Activity in Aging Mice
Human adipose tissue-derived mesenchymal stem cells (ADMSCs) were intravenously or intracerebroventricularly transplanted into the brains of 18-month-old mice once or four times at two-week intervals. Transplantation of ADMSCs improved both locomotor activity and cognitive function in the aged animals, in parallel with recovery of acetylcholine levels in brain tissues. [J Neurosci Res] Abstract
